"Adaptive Clinical Trials: An Introduction"
I provide an overview of adaptive clinical trials. After providing some background on what is an adaptive clinical trials and why one might want to do it, I focus on the "seamless Phase II/III trial" which is the most controversial application. An example illustrates some of the potential problems with the design, especially that the potential advantages of such a design often are illusory. Two methods for combining P values from the different phases of the seamless Phase II/III trial are presented, and some problems from an inferential point of view are discussed.
